Number 1 - the TEMPO Study
The best paper of 2012 [in my view] has to be the TEMPO study. Published in the NEJM, it puts a new class of drugs on the map for the treatment of polycystic kidney disease. It represents the end-result of decades of research by bench and clinical scientists. Of course, tolvaptan may be too expensive right now, and it's use may be limited by side-effects, and the long-term safety data isn't available, but really all of this will come - just wait! We have a therapy that actually seems to work in modifying an important kidney disease that, hitherto left untreated, leads to kidney failure.
